Expert started a clinical genetics lab at the Carolinas Medical Center. The lab made fee-for-service molecular diagnostics of cystic fibrosis and several other genetic diseases. He also manufactured fluorescent probes for cytogenetics. He was the director of new assay development at the Genzyme Genetics DNA Lab, and improved processes for DNA extraction and quality control.
Expert has wide experience with DNA and applications. He has made extensive sequencing, cloning and expression experiments. He has synthesized DNA with ABI and other instruments. He is an expert on DNA hybridization kinetics, and DNA sequencing by fluorescence and chemical cleavage methods. He is also an expert in mass spectrometric analysis of DNA, and has a passing experience with NMR analysis of DNA. Expert developed several probe technologies and holds two patents on processes in the field. In addition, he developed a novel method for heteroduplex analysis called a heteroduplex generator, for which he received over a million dollars in federal and state grant funding. He has developed probes for human papillomavirus, human genetic diseases (including cystic fibrosis, Factor V leiden, and Bloom's Syndrome). Also, Expert developed a novel method for rapid genotyping using MALDI-TOF MS.
Expert was part of a team in Germany that first described the human cytomegalovirus transcription enhancer element. Expert did primary sequencing and developed a gel shift assay to identify factors associated with the enhancer element. He also discovered other elements in the virus DNA associated with initiation of transcription by RNA polymerase III using a run-off type assay.
Expert holds a Ph.D. in medical microbiology and a Master's in applied microbiology. He has extensive research experience with E. coli, bacteriophage phi X 174, the Epstein-Barr virus, human cytomegalovirus, human papillomavirus and bacteriophage T4. He designed DNA tests for all these organisms plus expression cloning in Pichia pastoralis. He has experience in electron microscopy, DNA hybridization, cloning, PCR, RNA extraction, mass spec, and DNA sequencing. In addition, he has some experience with antibody production and protein analysis.
Expert developed several genotyping methods, and has experience in the practice of high through-put genotyping. Expert pioneered a method using heteroduplex generator, and received more than a million dollars in grant funding to make a large scale study genotyping patient DNA in the Rh blood group locus. Expert was the Director of Quality Control for the MASDA genotyping assay at the Genzyme Genetics DNA lab. Over 2000 patient samples a month were processed for up to 100 genotypes each. Expert also helped to provide genotyping data of the human papillomavirus in several large studies.
Expert is an expert in the genetics of the Epstein Barr virus, human cytomegalovirus human papillomavirus, and bacteriophages phi-x-174 and T4. He has several primary publications in these areas. His work was essential in elaborating the genotype/phenotype relationships in EBV regarding the natural history of the virus. Expert worked with the team that discovered and characterized the enhancer element of hCMV. Expert worked with several groups to help define type specific differences in early antigen expression in human papillomavirus.
Expert recently made a sabbatical year with the Richert Research group at Tufts University. During this time, he learned essential skills for MALDI-TOF MS in the context of nucleic acid and protein analysis. His research work on high-throughput genotyping was published in Nature Biotechnology. He currently serves as the Senior Genomics Scientist at Bruker Daltonics, and has experience in the analysis of nucleic acids and proteins by ion trap and electrospray methods in addition to MALDI. He has several publications in regard to this work.
